Current treatments for chronic hepatitis B (CHB) are associated with low rates of cure, making functional cure a viable treatment endpoint. The review discusses the progress in RNAi therapeutics, including early phase clinical trials that show promising safety data and significant reductions in HBsAg levels. However, challenges remain, particularly regarding the limitations of current RNAi therapies in targeting HBsAg derived from integrated hepatitis B DNA. The review emphasizes the need for combination therapies to achieve a functional cure.
